用流式细胞仪测定草的DNA含量

这几天一直给别人做FCM，测定几种草中的DNA含量（pg/2C），一开始还是很明白的，可是随着实验前进，慢慢我也糊涂了，两次的实验结果完全相反，我都不知道哪里错了。重新配置了所有药品，结果都和第一次不一样，可是只有第一次里面有我要的数据啊？难道传说中的偶然？
没办法，找来文章看，确无意间发现，crop science society of amercia期刊居然可以免费下载全文，而且还是SCI索引源之一呢。详见http://crop.scijournals.org/?ck=nck和http://www.scijournals.org/。呵呵，希望这样免费的地方越来越多。
这次测定主要的方法来自”Determination of the level of variation in polyploidy among Kentucky Bluegrass Cultivars by Means of Flow Cytometry“.
Introduce FCM–Flow cytometer
Flow cytometry is an analytical technique that can be used to study many different aspects of cytology and genetics. For expample, it can be use to study the cell cycle by determining the proportions of cell in G1,S,and G2/M stages of the cell cycle, thus allowing the calulation of cell cycle times(Marie and Brown, 1993). It is also useful in performing task such as chromosome sorting and quantification of DNA content. Thus, flow cytometry enables a quick estimation of DNA amount by quantifying nuclear DNA content. Flow cytometry has the advantage fo being faster and less labor intensive than other methods of DNA content estimation, such as the microdensitometer technique and cytological chromosome counts.

碎片，要看是什么碎片。我做的时候，一开始也是碎片过多，仔细观察发现，碎片分为两种，一种是植物纤维细胞，一种是要测定的植物细胞。哦，临时有事情，如果希望继续探讨，给我发email，告诉我你测定的是什么草，采用的那种前处理方法及所用的机器幸好。呵呵，我也不是什么高人，就是多做了一些试验。
我测定的是松树，算了一下我用过的前处理方法有20多种了，包括LB01，Tris-MgCl2，WPB，NIB，Gailbrainth’s等等，查到的方法都试过了，总之所有的方法都使细胞破碎了。你是怎么处理得到完整细胞核的？请问你的email 地址是？我得email是zhxy2008bj@163.com，用的机器具体型号我也不知道，不过前两年有人就用那机器做出来了。多谢了！
